I went through the setup all correctly but when i run the shortcut it give me an error

"Playback failed, possibly due to an invalid or conflictying IRQ"

Before i done this i could play the game but the sound kept breaking up, but now i cant run it at all i just keep geting that.

Can anyone help?

I've only ever seen that come from having the Music enabled. Try running set up and choosing no music. It sucks, I know, but it might be a way to get the game to run with at least one part of the audio; until someone else smarter than both of us figures something out :|

Try this out goto the Duke3D directory and rightclick on the SETUP.EXE and choose "run with VDMS" and when you setup your sound config it will detect the right sound config for Duke3D but don't test the sound, this is if you are using the VDMS soundblaster emulator, then right click DUKE3D.EXE and choose "run with VDMS" and see if the game will load with sound working properly.

hope this works for ya :)
